The combination of PCR Buffer without MgCl
2 and the MgCl
2 Stock Solution buffer are optimized for the amplification of specific DNA fragments in conventional PCRs (
i.e., with Taq DNA Polymerase). However, the MgCl
2 Stock Solution may be used with other, more appropriate PCR buffers to optimize any type of PCR (
e.g., hot start PCR, high fidelity PCR, long template PCR).
Allgemeine Beschreibung
The MgCl
2 Stock Solution is used for the individual adjustment of the Mg
2+-concentration in the PCR reaction and can be used in combination with the PCR buffer without MgCl
2. In most applications a concentration of 1.5mM MgCl
2 will yield satisfactory results with a dNTP concentration of 200μM each. In many cases, however, it is required to titrate the optimal Mg
2+ concentration to increase the specificity and the yield of the PCR reaction.
