Allgemeine Beschreibung
The TTC8 gene is also called as BBS8 and maps to human chromosome 14q32.11 with a span of 15 exons. It comprises of several tetratricopeptide repeats (TPRs) that function in protein–protein interactions. It interacts with PCM-1, a factor involved in basal body function. It also associates with centriolar structures.
Biochem/physiol Actions
TTC8 (tetratricopeptide repeat domain 8) gene encodes a centrosomal and basal body protein, a core component of BBSome, a complex that functions in the sorting of membrane proteins to primary cilia. This complex is essential for ciliogenesis. The complex is involved in the recognition of ciliary targeting signals and assembling a polymerized coat in order to direct membrane protein clusters to the ciliary membrane. Mutations in this gene have been linked to nonsyndromic retinitis pigmentosa that is characterized by night vision blindness and loss of midperipheral visual field. Defects in this gene are also associated with Bardet-Biedl syndrome, characterized by retinal dystrophy, obesity, polydactyly, renal malformations and learning disabilities.
