Reaction Conditions:
Stage #1: chloroacetonitrile;benzylaminewith potassium carbonate;sodium iodide in acetonitrile at 80; for 18 h;
Stage #2: with hydrogenchloride in diethyl ether;
Steps:
2.5 4.2.1.1 Synthesis of amino-acetonitrile hydrochlorides 2 and 3
General procedure: To a solution of chloroacetonitrile (1.1 equiv) in acetonitrile was added the primary amine (1 equiv), potassium carbonate (3 equiv) and sodium iodide (1 mol %). The reaction mixture was stirred at 80 °C for 18 h after which the precipitate was filtered and mother liquor concentrated in vacuo to yield a dark oil. The amino acetonitrile was dissolved in diethylether and treated with ethereal 1 N HCl (excess) to yield the hydrochloride salt, which was filtered and dried under vacuum to yield the desired product.
