Under nitrogen protection, 50 mL of acetone, 10.0 g (0.018 mol) of compound II, and 2.0 g of potassium fluoride were added to the reaction flask, and the reaction was incubated at 20°C, and TLC monitoring showed that there was no compound of formula II. The reaction solution was diluted into 1000 mL of ice water, stirred for 1 hour, filtered, washed with water, and dried at 60-70° C. for 20 hours to obtain 8.9 g of the compound of formula III (betamethasone propionate) with a yield of 99.2% and HPLC purity of 99.0%.
